Following Bury's penalty loss to Stoke City at the bet365 Stadium on Tuesday evening, City are now guaranteed a place in the knock-out stages of this season's competition.
A point, or a loss by less than a two-goal margin, at Morecambe's Globe Arena would confirm City as winners of Northern Group C, securing a home draw in round two in the process.
After leading City to victories against Stoke City and Bury in their previous Checkatrade Trophy group matches, Black will once again take charge of the side at Morecambe.
Black said: "This Wednesday's game is certainly an opportunity for some players to stake a claim for a future starting slot.
"We were naturally disappointed with Saturday's result (vs Accrington Stanley) but there is nothing we can do about that now.
"As much as it pains us to be out of the (FA) cup at the first hurdle, we want to concentrate now on making progress in this competition (Checkatrade Trophy).
"Ultimately, you can get to Wembley in this competition and win a trophy. Obviously that is a long way off yet but we will certainly be sending the right signals out to the players.
"Wednesday is an opportunity for us to get back to winning ways before our league game (vs Rochdale) on Saturday."
